چکیده

dae larvae and has been known as a traditional medicine in China. Many studies have shown that CS possesses anti-fatigue, antitumor, antioxidant, immunomodulatory, hypoglycemic, and vasorelaxant activities. Since a relationship between depression and fatigue has been well established, we determined to evaluate its antidepressive effect using the tail suspension test (TST) that, in parallel with the forced swimming test, has been proposed as a primary screening test for antidepressant drugs. The most prevalent theory for the pathogenesis of depression is the “monoamine hypothesis” in which monoamines (e.g., norepinephrine, dopamine, and serotonin) are thought to play important roles in the development of symptoms. Many antidepressant drugs work by modulating these neurotransmission systems. Recently, behavioral tests have been used to study mechanisms underlying some antidepressant-like substances with inhibitors of neurotransmitters such as alpha-1 adrenoreceptor antagonists, selective dopamine D2 receptor antagonists, and serotonin synthesis inhibitors. In addition to these screening behavioral tests of antidepressants, counting the numbers of head twitches induced by 5-hydroxytryptophan in mice is an effective method to evaluate serotoninerigic effects of drugs in vivo. It has been generally accepted that numbers of head twitches represent the level of 5-hydroxytryptamine (5-HT) in the synapses. In the present study, we have evaluated the antidepressantlike effects of hot water extract (HWCS) and supercritical fluid extract (SCCS), obtained from CS, using the TST in mice. We have also dissected the mechanism underlying the antidepressant actions of SCCS by examining the effects of pretreatment with an alpha-1 adrenoreceptor antagonist, a selective dopamine D2 receptor antagonist, and an inhibitor of serotonin synthesis. Furthermore, to determine whether the antidepressive effect of SCCS is mediated by the serotoninergic system, we have evaluated the effect of SCCS on 5-HTPinduced head twitch response in mice.

In eukaryotic cells, combinatorial phosphorylation of the hydroxyl residues on the inositol ring of phosphatidylinositol (PtdIns) gives rise to seven phosphoinositides (eight if PtdIns itself is included) (Fig. 1). In the ‘canonical’ phosphoinositide (PI) cycles, phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] serves as a precursor of the intracellular second The Physiology of Phosphoinos...

Biocides have a wide variety of uses, and their concentrations and exposure times vary according to usage. Recently, MRSA isolates with decreased biocide susceptibilities have been isolated from clinical samples, and MRSA isolates carrying antiseptic-r...

of dihydropyrazines (DHPs) such as 2,3-dihydro-5,6-dimethylpyrazine (Me-DHP), which causes DNA strand breakage in the plasmid pBR322. We showed that DHPs inhibit growth and cause mutagenesis in Escherichia coli and that Me-DHP is cytotoxic and genotoxic in E. coli due to the production of radical species in the presence of Cu . Phenyl substituted dihydropyrazines (Ph-DHPs) have much higher DNA ...

hibitors used to treat human immunodeficiency virus (HIV)1. Tenofovir can be administered once-daily at a dose of 300 mg in combination with other antiretroviral drugs. The advantage of once-daily dosing is a reduction in patient drug load. Also, tenofovir appeared to be well tolerated with low rates of drug-related therapy discontinuation in several clinical trials. Thus, tenofovir use is expe...
